DNA Topoisomerases: Single Gyrase Caught in the Act
نویسنده
چکیده
A recent study has analysed the action of bacterial DNA gyrase on a single substrate DNA molecule, discriminating the initial DNA wrapping and subsequent supercoiling steps in the reaction cycle.

Detection of DNA Gyrase Mutation and Multidrug Efflux Pumps Hyperactivity in Ciprofloxacin Resistant Clinical Isolates of Pseudomonas aeruginosa
Target modification and reduced drug accumulation are the main resistance mechanisms against fluoroquinolone antibiotics in Pseudomonas aeruginosa. We performed a genotypic characterization of three major Mex multidrug efflux pumps (MexAB-OprM, MexXY-OprM and MexCD-OprJ) in ciprofloxacin resistant clinical isolates of P. aeruginosa, collected from Tehran, Iran this was followed by sequencin...
متن کاملBacterial topoisomerases, anti-topoisomerases, and anti-topoisomerase resistance.
Topoisomerases are ubiquitous enzymes necessary for controlling the interlinking and twisting of DNA molecules. Among the four topoisomerases identified in eubacteria, two, DNA gyrase and topoisomerase IV have been exploited by nature and the pharmaceutical industry as antibacterial targets. Natural products that are inhibitors of one or both of these topoisomerases include the coumarin and cyc...
متن کاملArchaebacterial reverse gyrase cleavage-site specificity is similar to that of eubacterial DNA topoisomerases I.
ATP-dependent type I topoisomerases from extremely thermophilic archaebacteria--reverse gyrases--drive positive supercoiling of DNA. We showed that reverse gyrase from Desulfurococcus amylolyticus breaks the DNA at specific sites and covalently binds to the 5' end. In 30 out of 31 sites located in pBR322 DNA fragments, cleavage occurs at the sequence 5'---CNNN/---(N is any base). The same rule ...
متن کاملDNA gyrase with a single catalytic tyrosine can catalyze DNA supercoiling by a nicking-closing mechanism
The topological state of DNA is important for replication, recombination and transcription, and is regulated in vivo by DNA topoisomerases. Gyrase introduces negative supercoils into DNA at the expense of ATP hydrolysis. It is the accepted view that gyrase achieves supercoiling by a strand passage mechanism, in which double-stranded DNA is cleaved, and a second double-stranded segment is passed...
متن کاملReverse gyrase—recent advances and current mechanistic understanding of positive DNA supercoiling
Reverse gyrases are topoisomerases that introduce positive supercoils into DNA in an ATP-dependent reaction. They consist of a helicase domain and a topoisomerase domain that closely cooperate in catalysis. The mechanism of the functional cooperation of these domains has remained elusive. Recent studies have shown that the helicase domain is a nucleotide-regulated conformational switch that alt...
